Product Specific Information
The B21.5 monoclonal antibody reacts with the Vβ10 T Cell Receptor (TCR) in mice of the b haplotype of the TCR Vβ gene complex, including AKR, BALB/c, CBA, C3H/He, C57BL, DBA/1, and DBA/2 mice.
It does not react with Vβ10 in mice of the a or c haplotypes, including C57BR, C57L, SJL, and SWR mice.
Applications Reported:
This B21.5 antibody has been reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of C57Bl/6 mouse splenocytes.
Recommended use ≤ 0.25 µg per test (100 µL final volume).
Cell number should be determined empirically (10⁵–10⁸ cells/test).
Careful titration is recommended for optimal performance.
Target Information
T cell receptors (TCRs) discriminate foreign from self-peptides presented by MHC class II molecules, which is critical for adaptive immune response.
Recognition of self-peptides has been linked to autoimmune diseases, and mutant self-peptides are associated with tumors.
Superantigens engaging TCRs can cause toxic shock syndrome.
Autoantibodies to Vβ segments have been found in patients with rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially blocking TH1-mediated inflammatory responses.
T Cell and TCR Diversity:
Most human T cells express TCR αβ and either CD4 or CD8 (single positive).
A small subset lacks both markers (double negative, DN), which may increase in autoimmune or immunodeficiency disorders.
γδ T cells, mainly in epithelial tissues, show less TCR diversity and exhibit antitumor and immunoregulatory activity.
For Research Use Only. Not for use in diagnostic procedures. Not for resale without express authorization.
