Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low but play a critical role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, triggering histamine release. IgE is expressed both as a soluble and membrane-bound form, and IgE21 recognizes both.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test (in 100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.
• Always use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

IgE is one of five immunoglobulin classes in mammals, consisting of two epsilon heavy chains and two light chains. It plays a central role in type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation, releasing mediators responsible for allergic symptoms. IgE also contributes to parasite immunity.
