Product Specific Information

Description:
The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE levels in serum are low and play a key role in allergic responses by binding to mast cells and basophils via Fc epsilon receptor I, leading to histamine release. IgE is produced by terminally differentiated plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Validated for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates use Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
Excitation: 552 nm; Emission: 663 nm; Laser: 561 nm (Yellow).

Target Information

IgE (Immunoglobulin E):
One of the five immunoglobulin classes in mammals. Composed of two epsilon heavy chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding Fc receptors on basophils and mast cells. Crosslinking induces degranulation, releasing histamine and other mediators. Also essential for antiparasitic immunity.
