Product Specific Information

The monoclonal antibody IgE21 recognizes the immunoglobulin E (IgE) class of antibodies. IgE is present at low levels in serum and plays a key role in allergic responses by binding to mast cells and basophils through the Fc epsilon receptor I, triggering release of histamine and other mediators. IgE is produced by plasma B cells and basophils and exists in both soluble and membrane-bound forms. IgE21 recognizes both forms.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved staining performance. Add 5 µL per sample to antibody cocktails prior to mixing with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures that enable engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

IgE is one of five classes of mammalian immunoglobulins. It consists of two heavy (epsilon) chains and two light chains. IgE mediates type I hypersensitivity reactions (e.g., hay fever, asthma, hives, anaphylaxis) by binding to Fc receptors on basophils and mast cells. Crosslinking of IgE induces degranulation and release of histamine and other molecules, contributing to allergic responses. IgE also plays a role in immune defense against parasites.
