Specificity Comments:
This monoclonal antibody recognizes a protein of approximately 50 kDa, identified as Glial Fibrillary Acidic Protein (GFAP). It shows no cross-reaction with other intermediate filament proteins. GFAP is specifically found in astroglia and is a well-established marker for benign astrocyte and neoplastic cells of glial origin in the central nervous system.
Antibody to GFAP is useful for differentiating primary gliomas from metastatic brain lesions and for documenting astrocytic differentiation in tumors outside the CNS.
Target Information
GFAP (Glial fibrillary acidic protein) is a member of the class III intermediate filament protein family. It is heavily and specifically expressed in astrocytes and certain astroglia of the central nervous system, satellite cells of peripheral ganglia, and non-myelinating Schwann cells of peripheral nerves. Neural stem cells also strongly express GFAP.
Antibodies to GFAP are widely used as markers of astrocytic cells. Many types of brain tumors derived from astrocytic cells express GFAP. It is also found in the lens epithelium, Kupffer cells of the liver, salivary tumor cells, and sometimes in erythrocytes. GFAP is used to distinguish astrocytes from other glial cells during development.
Mutations in the GFAP gene cause Alexander disease, a rare disorder of astrocytes in the CNS. Alternative splicing results in multiple transcript variants encoding distinct isoforms.
