Target Information

ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1, is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities, playing a crucial role in ER-based stress signaling.
ERN1 controls IRE1 proteolysis in mammalian cells and is essential for fetal viability. Specifically, IRE1 acts as a sensor of unfolded proteins in the ER and initiates the unfolded protein response by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
Null mutants are viable but IRE1 is essential under stress conditions that cause unfolded protein accumulation in the ER. Depletion of IRE1p via dominant negative expression leads to apoptosis under such stress conditions.
