Target Information
ERN1 (endoplasmic reticulum to nucleus signaling 1) is a serine/threonine kinase that functions as a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in ER-based stress signaling.
ERN1 regulates IRE1 proteolysis in mammalian cells and is essential for fetal viability in extraembryonic cells.
IRE1 acts as a sensor of unfolded proteins in the ER, transmitting stress signals to the nucleus by splicing XBP1 mRNA into a potent unfolded-protein response transcriptional activator.
Null mutants are viable, but IRE1 is critical under stress conditions that cause unfolded protein accumulation in the ER.
Depletion of IRE1p has minimal effects under normal conditions but induces apoptosis under ER stress.
