• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, then cool at RT for 20 min.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker and may function as a surface receptor involved in adhesion, differentiation, and proliferation of hematopoietic stem cells. CD34 expression reflects specific developmental stages and may mediate attachment to bone marrow extracellular matrix or stromal cells. It can act as a scaffold for lineage-specific glycans, facilitating interactions with lectins or selectins. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
