Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature (RT).
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at RT for 20 minutes.
Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker with reversible expression on human hematopoietic stem cells. CD34 may function as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells.
CD34 expression reflects specific developmental states of hematopoietic cells with distinct adhesion and differentiation properties. It may act as an adhesion molecule mediating stem cell attachment to bone marrow extracellular matrix or stromal cells, and as a scaffold for binding lineage-specific glycans, facilitating interactions with lectins on stromal or marrow cells. The intracellular domain can be phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
