• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker, with reversible expression on human hematopoietic stem cells. CD34 may function as a surface receptor involved in adhesion, differentiation, and proliferation of stem and progenitor cells.

It is likely involved in hematopoietic development, mediating attachment of stem cells to bone marrow extracellular matrix or stromal cells. CD34 may act as a scaffold for lineage-specific glycans, enabling binding to lectins expressed by stromal or marrow components. It also presents carbohydrate ligands to selectins, and its intracellular domain can be phosphorylated by activated protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
