Product Specific Information

The KGP130 monoclonal antibody reacts with mouse gp130 (CD130), a 130 kD Type-I transmembrane glycoprotein. It consists of a 597 amino acid extracellular domain, a single transmembrane domain, and a 277 amino acid cytoplasmic domain. gp130 forms part of multiple heterodimeric receptors, including those for IL-6, IL-11, IL-27, Oncostatin M, and LIF. A soluble form of gp130 can inhibit IL-6 signaling.

gp130 is mainly expressed on T cells, monocytes, endothelial cells, activated B cells, and plasma cells, with lower expression on most leukocytes and epithelial cells. Upon ligand binding, gp130 undergoes tyrosine phosphorylation, activating PI3 kinase, Ras-MAPK, and Stat pathways.

Preincubation of mouse splenic T cells with IL-6 can block KGP130 binding to gp130, suggesting the binding site overlaps the IL-6 interaction domain. No reactivity has been observed with human gp130.

Target Information

The encoded protein acts as a signal transducer shared by several cytokines, including IL-6, CNTF, LIF, and OSM. It functions as part of a cytokine receptor complex, with activation dependent on cytokine binding. Viral IL6 (vIL6) from Kaposi sarcoma-associated herpesvirus can activate this protein directly. Mouse knockout studies suggest a key role in regulating myocyte apoptosis. Multiple alternatively spliced transcript variants exist.
