Product Specific Information

The monoclonal antibody MPO455-8E6 recognizes myeloperoxidase (MPO), a protein within the azurophilic granules of myeloid cells. MPO consists of two 55 kDa and two 15 kDa subunits; the larger subunits contain a heme group giving a greenish color. As an enzyme, MPO decomposes hydrogen peroxide and oxidizes tyrosine, producing hypochlorous acid and tyrosyl radicals responsible for neutrophil cytotoxic activity. MPO is clinically relevant for cancer diagnosis and correlates with cardiac events.

Applications Reported:
This antibody is used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
For optimal results:

1. Stain whole blood with surface antigen antibodies.
2. Treat with 1X RBC lysis buffer (Product # 00-4333-57).
3. Fix cells with IC Fixation Buffer (Product # 00-8222-49).
4. Wash twice with Permeabilization Buffer (Product # 00-8333-56).
5. Incubate with MPO455-8E6 for 30–60 minutes.

After washing, analyze on a flow cytometer. Use 5 µL (0.5 µg) per test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test.

Optical Properties:
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation and is a major component of neutrophil azurophilic granules. It forms a tetramer of two light and two heavy chains. MPO generates hypohalous acids essential for neutrophil microbicidal activity. Associated diseases include Myeloperoxidase Deficiency and Familial Alzheimer’s Disease type 1.
