Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified on acute lymphoblastic leukemias (ALL). CD10 is identical to neutral endopeptidase (NEP) and acts as a Zn²⁺-dependent metallo-peptidase, regulating chemotactic and inflammatory processes involving neutrophils. It also influences stromal cell-dependent B lymphopoiesis and is expressed in germinal center B cells, enterocytes, and renal tubules.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
    • Recommended use: 5 µL (0.125 µg) per 100 µL test volume
    • Cell range: 10⁵–10⁸ cells/test

Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emitting at 702 nm. Use a 710/50 bandpass filter for detection.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark.
Excitation: 405 nm
Emission: 702 nm
Laser: Violet
Filtration: 0.2 µm post-manufacturing filtered

Super Bright Polymer Dyes are sold under license from Becton, Dickinson and Company.

Target Information

CD10 (CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed in lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and chronic myelocytic leukemia (CML) cells. CD10 is also present in breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed in kidney and intestinal epithelial brush borders.

It cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones such as glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. The CD10 gene produces four mRNA transcripts due to alternative splicing of the 5′ untranslated region.

Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
