Product Specific Information
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8 alpha/beta heterodimer or CD8 alpha/alpha homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with p56lck, playing a role in T-cell development and activation.

- Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells (5 µL or 0.06 µg/test)
- Super Bright 702: Tandem dye excited at 405 nm, emits at 702 nm (recommended 710/50 bandpass filter)
- Staining Panels: Use Super Bright Staining Buffer (SB-4400) to minimize polymer interactions
- Light Sensitivity: Protect from light due to photo-induced oxidation
- Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal impact on brightness

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein involved in signal transduction and T cell differentiation. CD8 is expressed on T cells as a CD8α/CD8β heterodimer, functioning as a co-receptor for MHC class I molecules. It also exists as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Interaction with MHC-I/peptide complexes recruits Lck kinase, leading to CTL activation and pathogen/tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
