Target Information
Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome particle. In eukaryotes, DNA double-strand breaks (DSBs) trigger phosphorylation of serine 139 at the carboxy terminus of H2AX, resulting in γ-H2AX.
Phosphorylation can be detected by Western blot or immunofluorescence, indicating DSB frequency.
Phosphatidylinositol 3-kinases are implicated in H2AX phosphorylation, though ATM, DNA-PK, and ATR may also contribute.
H2A.X contains 143 amino acids, synthesized in G1 and S-phase. Its mRNA exists in two forms (polyA+ and polyA−), corresponding to basal and replication-type histone mRNAs.
Histone H2A.X maps to human chromosome region 11q23.2–q23.3 and contributes to DNA structure and repair, regulating MDC1 interaction at DNA damage sites.
