Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic region has tyrosine phosphatase activity, critical for lymphocyte activation.

Applications Tested: Pre-titrated for flow cytometry; use 5 µL (0.25 µg) per 100 µL test volume. Suitable for 10⁵–10⁸ cells/test.

Fluorophore Information:
PE-eFluor® 610 is excited at 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Handling and Storage:

• Light sensitive; protect from light.
• Store at 4°C, do not freeze.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333).
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, representing ~10% of lymphocyte surface proteins. It regulates Src-family kinases and is essential for T- and B-cell receptor signaling. CD45 isoforms arise from alternative splicing and differ in extracellular domains. CD45 also modulates integrin-mediated signaling, apoptosis, and cytokine receptor pathways.
