Product Specific Information

Description:
FA-11 monoclonal antibody reacts with mouse CD68 (macrosialin), a heavily O- and N-glycosylated type I integral membrane protein of the sialomucin family. CD68 is mainly localized in late endosomes and lysosomes, with a smaller fraction at the cell surface. It serves as a pan-macrophage marker but can also be expressed in Langerhans and dendritic cells. CD68 binds oxidized LDL and participates in phagocytosis.

Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated for intracellular staining of mouse bone marrow-derived macrophages using Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤1.0 µg/test (100 µL final volume). Optimize empirically for best performance.

Fluorochrome Information:
PE-eFluor 610 is excitable by 488–561 nm laser lines and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent). Ensure instrument compatibility.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) up to 3 days at 4°C in dark with minimal brightness loss.

Excitation / Emission / Laser:
488–561 nm excitation; 607 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Target Information

CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein mainly expressed in lysosomes of monocytes and macrophages, and to a lesser extent in dendritic cells and granulocytes. It plays roles in phagocytosis and cell-pathogen interactions. CD68 expression is elevated in certain malignancies, including AML and histiocytic disorders.
