Product Specific Information

Description:
The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorophore Information:
PE-eFluor 610 can be excited with 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (equivalent to PE-Texas Red). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm / 607 nm / Blue, Green, Yellow-Green lasers.

Target Information

Human CD62L is a 74–95 kDa glycoprotein adhesion receptor (L-Selectin) consisting of:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

It is constitutively expressed on most leukocytes including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
