Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, a transmembrane and cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, peripheral and central nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R modulates macrophage activation, inhibits mast cell degranulation, and reduces TNFα and IL-13 secretion, contributing to immune regulation. CD200 expression on trophoblasts suggests a role in maternal tolerance.

Applications Tested: Tested on normal human peripheral blood cells. Recommended ≤ 0.06 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize titration for best results.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) improves staining performance. Add 5 µL per sample (regardless of panel size) prior to mixing with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescence and minimal spillover.

Target Information

CD200 is a type-1 membrane glycoprotein with two Ig domains, part of the immunoglobulin superfamily. It interacts with CD200R on myeloid cells to inhibit macrophage activation. Homologues exist in herpesviruses and poxviruses. CD200 regulates myeloid activity and delivers inhibitory signals to macrophages. Multiple splice variants exist. Associated diseases include Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy.
