Product Specific Information

The monoclonal antibody OX104 recognizes human CD200 (OX2), a member of the Ig superfamily with two Ig domains, a transmembrane, and a cytoplasmic domain. CD200 is expressed on resting and activated B cells, subsets of T cells, keratinocytes, nerve cells, follicular dendritic cells, and ovarian cells. Interaction with CD200R modulates immune responses by reducing macrophage activation, mast cell degranulation, and cytokine secretion.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis.

Tested Applications

• Tested by flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: ≤5 µL (1.0 µg)/test.
• One test = amount of antibody for 100 µL cell sample (10⁵–10⁸ cells/test).
• Optimize antibody titration for best performance.

Important Notes

• NovaFluor dyes are not compatible with DNA-intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of blocking buffer per sample or per 100 µL of cell suspension.

Target Information

CD200 is a type-I membrane glycoprotein in the immunoglobulin superfamily with two Ig domains. It interacts with CD200R on myeloid cells to inhibit macrophage activation. CD200 homologues exist in several herpesviruses and poxviruses. Dysregulation of CD200 is associated with diseases such as Mantle Cell Lymphoma and Primary Hypertrophic Osteoarthropathy. Multiple alternatively spliced variants exist.
