This antibody reacts with SLP76, a 76 kDa cytosolic adaptor protein involved in signaling of various hematopoietic cells such as T cells, mast cells, and neutrophils. In B cells, it is replaced by SLP65.

Target Information

SLP-76 was originally identified as a substrate of the ZAP-70 protein tyrosine kinase following T cell receptor (TCR) ligation in the leukemic T cell line Jurkat.
The SLP-76 locus is located on human chromosome 5q33, and both human and murine cDNAs encode 533 amino acid proteins that are 72% identical.
The protein comprises three modular domains:

• NH2-terminus: acidic region with a PEST domain and tyrosine residues phosphorylated upon TCR ligation
• Central region: proline-rich domain
• COOH-terminus: SH2 domain

SLP-76 associates with multiple proteins both constitutively and inducibly following receptor ligation, functioning as an adaptor or scaffold protein.
Studies using SLP-76 deficient T cell lines or mice show its role in promoting T cell development, activation, mast cell, and platelet function.
