Product Specific Information
The MTS510 monoclonal antibody reacts with the mouse Toll-like receptor 4 (TLR4)/MD-2 complex. TLR family members are type I transmembrane proteins characterized by extracellular leucine-rich repeats and a cytoplasmic domain homologous to the IL-1 receptor. TLR4 associates with MD-2 and CD14 to form a complex responsible for LPS recognition and signaling.
In mice, TLR4 is expressed by thioglycolate-elicited peritoneal macrophages and is downregulated upon LPS stimulation. The TLR4 gene is defective in C3H/HeJ and C57BL/10ScCr mice, which are hyporesponsive to LPS.
MTS510 co-immunoprecipitates MD-2 (~30 kDa) and TLR4 (~100 kDa), preferentially binding to TLR4 associated with MD-2. In contrast, UT41 binds TLR4 regardless of MD-2 association.
Applications Reported
- Flow cytometric analysis
- Immunoprecipitation
- Immunohistology of frozen tissue sections
- Inhibition of LPS-induced cytokine production
For functional assays, use Functional Grade purified MTS510 (Product #16-9924-81).
Applications Tested
Tested by flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells. Recommended ≤1 µg/test (in 100 µL final volume). Optimal titration recommended per assay.
Purity and Quality
- Purity: >90% (SDS-PAGE)
- Aggregation: <10% (HPLC)
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
MD-2 is a member of the Toll/interleukin-1 receptor (TIR) family, including Toll-like receptors (TLRs). TLRs recognize pathogen-associated molecular patterns (PAMPs) and link innate and adaptive immunity. TLR4, the major receptor for lipopolysaccharide (LPS), requires MD-2 binding for maximal response. The LPS response specificity depends on MD-2 species; for example, human MD-2 in mouse cells can alter TLR4 response to LPS analogs.
