The G-protein-coupled receptor (GPCR) superfamily includes approximately 600–1,000 members, representing the largest class of molecular targets with therapeutic relevance.
The nociceptin receptor is a G-protein coupled opioid receptor that binds the endogenous neuropeptide nociceptin. Ligand binding induces conformational changes that activate G protein signaling and modulate downstream effectors. This signaling inhibits adenylate cyclase and calcium channel activity. Arrestins regulate G protein signaling and activate MAP kinase pathways.
The receptor plays roles in nociception, pain perception, and regulation of locomotor activity by nociceptin.
