Target Information

Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis, a cell death process removing toxic or useless cells during mammalian development, can be induced by various stimuli, including death factors of the tumor necrosis factor family, UV irradiation, anticancer drugs, and deprivation of survival factors. The apoptotic process involves shrinkage and fragmentation of cells and nuclei, and degradation of chromosomal DNA into nucleosomal units.

CAD, activated by caspases, exists as a complex with its inhibitor, ICAD (inhibitor of CAD). ICAD assists in proper folding of CAD and inhibits its DNase activity. During apoptosis, activated caspases cleave ICAD, releasing CAD to enter the nucleus and cleave chromosomal DNA. Caspase activation is controlled by the balance between pro- and anti-apoptotic proteins of the Bcl-2 family, heat shock proteins, and inhibitors of apoptosis proteins (IAPs). Fourteen members of the caspase family have been identified, seven of which mediate apoptosis.
