Target Information
ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis by inhibiting caspase-3-activated DNase (CAD). It exists in two isoforms:
- ICAD-L/DFF45 (45 kDa): functional isoform
- ICAD-S/DFF35 (35 kDa): shorter, non-functional isoform
Both isoforms can bind CAD, but only ICAD-L is functionally active. During apoptosis, ICAD is cleaved by caspase-3 at residues 117 and 224, releasing CAD to execute DNA fragmentation. ICAD also acts as a chaperone for CAD folding.
Recent studies suggest ICAD may serve as a potential therapeutic target for restoring normal apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified.
