Product Specific Information

Antibody detects endogenous levels of TPOR only when phosphorylated at Tyr626.

Target Information

In 1990, the oncogene v-mpl was identified from the murine myeloproliferative leukemia virus, capable of immortalizing bone marrow hematopoietic cells.
In 1992, the human homologue c-mpl was cloned, encoding a protein homologous to hematopoietic receptor superfamily members.
Anti-sense oligodeoxynucleotides of c-mpl inhibited megakaryocyte colony formation.
The ligand thrombopoietin (TPO), cloned in 1994, is the major regulator of megakaryocytopoiesis and platelet formation.
The c-mpl gene product (CD110) is a 635 amino acid transmembrane protein with extracellular cytokine receptor domains and intracellular receptor motifs.
TPO-R deficient mice are severely thrombocytopenic, highlighting the critical role of CD110 and thrombopoietin in platelet formation.
Upon thrombopoietin binding, CD110 dimerizes, activating JAK, STAT, MAPK, Shc, and receptor tyrosine phosphorylation pathways.
