Product Specific Information

The monoclonal antibody eBio84-3C1 reacts with CD43, also known as leukosialin, galactoglycoprotein, and sialophorin. CD43 is a sialomucin that can exhibit both adhesive and anti-adhesive functions. It is expressed on most leukocytes except resting B lymphocytes, and its surface expression decreases upon activation due to proteolytic processing. CD43 is involved in activation of T cells, B cells, NK cells, and monocytes. The counter-receptor for CD43 is CD169/SIGLEC-1, expressed on macrophages.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.4 µg) per test. A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which contains improved formulation over the original CellBlox. It is required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures that provide engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD43 (leukosialin, sialophorin) is a transmembrane mucin-like protein with a high negative charge, expressed on most hematopoietic cells. It forms a repulsive barrier that interferes with adhesion but can also promote leukocyte aggregation. Through interactions with actin-binding proteins (ezrin, moesin), CD43 regulates T-cell morphology and cell-cell interactions during lymphocyte traffic. CD43 signaling modulates LFA-1 adhesiveness and can both enhance and counteract LFA-1 induction. Expression of CD43 induces tumor suppressor p53 but may promote tumor proliferation in p53/ARF-deficient contexts. CD43 dysfunction is associated with Wiscott-Aldrich Syndrome and Adenoid Basal Cell Carcinoma.
