Product Specific Information

This BB7.2 monoclonal antibody recognizes the human MHC class I molecule HLA-A2, a member of the HLA-A family. MHC class I molecules are composed of a single alpha chain with three domains, associated with beta 2-microglobulin, and expressed on all nucleated cells.

The BB7.2 antibody exhibits blocking activity and also recognizes HLA-A28.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
Use 5 µL (0.6 µg) per test (defined as staining a 100 µL cell sample). Cell number may vary from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
• The conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which must be used for optimal staining.
  • Add 5 µL of CellBlox Plus per sample, regardless of the number of NovaFluors.
  • For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

HLA-A belongs to the HLA class I heavy chain family. It forms a heterodimer with beta-2 microglobulin.
These molecules present peptides from the endoplasmic reticulum lumen to the immune system and are expressed in nearly all cells.
The heavy chain (~45 kDa) gene has 8 exons encoding the leader peptide, peptide-binding domains (α1, α2), α3 domain, transmembrane region, and cytoplasmic tail.
Polymorphisms in exons 2 and 3 determine peptide-binding specificity, critical for transplantation typing.
HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding neoantigen identification via LC-MS/MS.
