Target Information

Estrogen Receptor beta (ERβ, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ERβ was initially cloned and characterized from testis.
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA.
ER can regulate gene transcription positively or negatively via recruitment of coactivator or corepressor proteins.
Two forms of estrogen receptor exist—alpha (ESR1) and beta (ESR2)—encoded by separate genes. ERβ binds estrogens with an affinity similar to ERα, and some ERβ isoforms dominantly inhibit ERα activity in reproductive tissues.
ERβ is widely expressed in various tissues and acts as a potent tumor suppressor, playing roles in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain Disease, and cardiovascular disease risk.
