Target Information
FOXO1 (FKHR, Forkhead Box O1)는 전사인자(forkhead homeotic gene family)의 하위 그룹 구성원으로, 핵 수용체 활성의 공동활성화자(coactivator) 또는 공동억제자(corepressor)로 작용할 수 있습니다.
PAX3와의 결합은 alveolar rhabdomyosarcoma와 관련이 있으며, FOXO1은 자가포식(autophagy), 근육 성장 및 분화 조절, 산화 스트레스에 대한 대사 항상성 유지에 관여합니다.
또한 FOXO1은 인슐린 반응 요소(IRE) 및 Daf-16 결합 요소(DBE)에 결합하여 유전자 발현을 조절하며, 골형성세포 수 및 골량 조절, 골격의 내분비 기능 조절에도 중요한 역할을 합니다.
간세포에서는 FOXO1이 PPARGC1A와 함께 작용하여 IGFBP1, G6PC, PPCK1 등의 발현을 촉진함으로써 포도당신생합성(gluconeogenesis)을 유도합니다.
FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 하류에서 세포사멸 조절에 관여하는 중요한 인자입니다.
