Target Information
Estrogen Receptor beta (ERβ, NR3A2) is a nuclear receptor protein of approximately 55 kDa, with isoforms ranging from 36–59 kDa. It was first cloned from human testis. ERs are ligand-activated transcription factors belonging to the steroid/thyroid hormone receptor superfamily. Upon estrogen binding, ERβ undergoes conformational change, dimerizes, and binds estrogen response elements (EREs) on DNA to regulate gene transcription via coactivator or corepressor recruitment.
Two main forms exist: ERα (ESR1) and ERβ (ESR2). ERβ binds estrogens with similar affinity to ERα, and some isoforms can inhibit ERα activity in reproductive tissues. ERβ is broadly expressed in human tissues and functions as a tumor suppressor, implicated in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
