SPHK1, along with SPHK2, catalyzes the phosphorylation of sphingosine to produce sphingosine-1-phosphate (S1P), a key signaling molecule.
S1P signals through G-protein coupled receptors and regulates diverse cellular processes such as:

  • Cell differentiation
  • Calcium mobilization from intracellular stores
  • Apoptosis

Inhibitors of SPHKs are considered potential therapeutics for cancer treatment.
S1P also interacts with EDG family G protein-coupled receptors (S1P Receptors).
