Product Specific Information

The monoclonal antibody 2D3 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and integrins (CD51/CD61, CD41/CD61). It mediates bi-directional signaling, regulates macrophage phagocytosis, and acts as a receptor for thrombospondin. CD47 is expressed in hematopoietic and non-hematopoietic cells.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.6 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample, regardless of the number of NovaFluors in the panel.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD47 (integrin-associated protein, IAP) is a cell surface transmembrane glycoprotein interacting with integrins and modulating signaling pathways. It functions in calcium regulation during cell adhesion, acts as a receptor for thrombospondin, and influences membrane transport and signal transduction. CD47 interacts with CD172a (SIRPα), inhibiting macrophage phagocytosis of CD47-positive cells. It also affects cell migration, B cell adhesion, T cell activation, chondrocyte response, and neuronal development.
