Product Specific Information

The monoclonal antibody 2D3 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP (signal regulator protein) and integrins CD51/CD61 and CD41/CD61. This interaction mediates bi-directional signaling affecting neural synaptic activity and macrophage phagocytosis. CD47 also acts as a receptor for thrombospondin.

Expression of CD47 is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and in non-hematopoietic cells. Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Tested

This antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended use: 5 µL (0.6 µg) per test in a final volume of 100 µL.
Cell number range: 10⁵–10⁸ cells/test.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls, use 5 µL of buffer per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD47 (Integrin-associated protein, IAP) is a widely expressed transmembrane glycoprotein that interacts with integrins and modulates signaling pathways, including those involving heterotrimeric G proteins. CD47 engagement increases intracellular calcium upon cell adhesion and acts as a receptor for thrombospondin. It plays roles in membrane transport, signal transduction, immune regulation, and neuronal development. The CD47–CD172a (SIRPα) interaction inhibits phagocytosis and influences cell migration, adhesion, and activation.
