Target Information
Nerve growth factor (NGF) influences the survival and differentiation of various neuronal and non-neuronal cells. NGF binds to specific high- and low-affinity receptors on the cell surface; only the high-affinity receptor mediates signal transduction. Cross-linking studies with ¹²⁵I-NGF show that the high-affinity receptor in rat PC12 cells forms a 158 kDa complex with NGF, while the low-affinity receptor forms a 100 kDa complex. Full-length cDNA clones for human and rat NGF receptor genes show greater than 90% homology. NGF promotes nerve growth and differentiation of sympathetic and sensory neurons and is involved in their regulation. Mutations in the NGF gene are associated with hereditary sensory and autonomic neuropathy type 5 (HSAN5), and dysregulation of NGF expression is linked to allergic rhinitis.
