Product Specific Information
This antibody recognizes a defined epitope (aa 16–25) on human p53, a 50 kDa intracellular tumor suppressor found in increased amounts in a wide variety of transformed cells. It is frequently mutated or inactivated in many types of cancer.
Recommended conditions: Western Blot – overnight incubation at 4°C under non-reducing conditions using SDS-PAGE (12% separating gel).
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates stress response genes such as p21^waf1^, leading to G1 or G2/M arrest.
The protein structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization. p53 functions as an anticancer factor by inhibiting proliferation of damaged cells. Mutations in TP53 are common in many cancers and can cause loss of tumor suppressor activity. Germline mutations are also linked to Li-Fraumeni syndrome.
