Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization. Phosphorylation at Ser392 is associated with tumor formation. p53 is linked to aging, oxidative stress, and learning/memory deficits. Normally expressed at low levels, it is upregulated in transformed cells, contributing to malignancy. Mutations in TP53 cause loss of tumor suppressor activity and occur in many cancers, including familial Li-Fraumeni syndrome.
