Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 regulates expression of genes such as p21waf1 involved in G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.
Phosphorylation at specific residues, including Ser15 and Ser392, plays key roles in activation, stabilization, and tumor association. Mutations in the TP53 gene are common in various cancers and can lead to loss of tumor suppressor activity. Germline mutations are also associated with conditions such as Li-Fraumeni syndrome.
p53 is typically present at low levels in normal cells but is upregulated in transformed or stressed cells, contributing to cell cycle regulation, apoptosis, and tumor suppression.
