Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for p53 activation and stabilization. p53 functions as an anticancer agent by inhibiting proliferation of damaged cells.
Mutations in p53, common in various cancers, disrupt DNA binding and tumor suppressor activity.
Alterations in the TP53 gene are associated with both somatic and germline mutations, including Li-Fraumeni syndrome.
p53 is also implicated in aging, oxidative stress, and cognitive processes such as long-term potentiation.
