Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 up-regulates stress response genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 correlates with tumor formation.
p53 plays a major role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also associated with aging, oxidative stress, and cognitive functions (LTP).
Mutations in the TP53 gene lead to loss of tumor suppressor activity and are found in many human cancers, including hereditary Li-Fraumeni syndrome.
