PGP9.5 (Protein Gene Product 9.5, UCH-L1, PARK5) is a neuron-specific protein distinct from neuron-specific enolase. It has a molecular weight of 27 kDa and belongs to the ubiquitin carboxyl-terminal hydrolase family (peptidase family C12).
PGP9.5 exhibits both ubiquitin hydrolase and ligase activities, generating ubiquitin monomers from small C-terminal adducts. It is expressed in:
- Neurons and nerve fibers (CNS and PNS)
- Neuroendocrine cells
- Renal tubules, spermatogonia, Leydig cells, and ova
- Pregnant and non-pregnant corpus luteum cells
Altered expression of PGP9.5 is associated with:
- Overexpression: non-small cell lung cancer
- Decreased expression: Huntington’s disease, Alzheimer’s disease
PGP9.5 serves as a neuronal marker and is valuable in neurodegenerative disease studies such as Parkinson’s disease.
