Product Specific Information

This SDRNR monoclonal antibody recognizes human and mouse AKT (Protein Kinase B, PKB) phosphorylated on Ser473.
AKT is a serine/threonine kinase involved in metabolism, proliferation, apoptosis/survival, and migration.
Phosphorylation at T308 (by PDK1) and S473 is required for full activation.
Activated AKT inhibits apoptosis via phosphorylation and inactivation of targets such as Bad, Foxo1, c-Raf, and caspase-9.
Deregulation of AKT contributes to various cancers.
Specificity of SDRNR clone was confirmed by ELISA, flow cytometry, and western blotting.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated; tested on normal human peripheral blood cells at 5 µL (0.06 µg)/test.
A test corresponds to staining of cells in a final volume of 100 µL (10⁵–10⁸ cells/test).


Staining Protocols

  • Protocol A: Two-step for cytoplasmic proteins; allows detection of surface and intracellular proteins.
  • Protocol B: One-step for nuclear proteins; suitable for transcription factors and phosphorylated proteins.
  • Protocol C: Two-step fixation/methanol; optimal for phospho-specific signaling discrimination.

Target Information

AKT1 (PKB alpha) is a serine/threonine kinase regulating cell survival.
Activated by insulin, PI3K, IGF1, and other growth factors, AKT inhibits apoptosis and promotes cell cycle progression via phosphorylation of multiple targets (e.g., forkhead transcription factors, caspase-9).
The AKT pathway is a major focus in cancer drug discovery.
