Product Specific Information
This CR3M monoclonal antibody reacts with human CCL3, also known as MIP-1 alpha (Macrophage Inflammatory Protein 1 alpha), a member of the CC chemokine family closely related to CCL4 (MIP-1 beta).
CCL3 plays a key role in leukocyte recruitment to inflammation sites, preferentially attracting CD8+ T cells, while CD4+ T cells respond more to CCL4. It induces inflammatory cytokine secretion, mast cell degranulation, and NK cell activation, and may inhibit hematopoietic stem cell proliferation to maintain quiescence.
CCL3 signaling occurs via CCR1, CCR4, and CCR5 receptors, which are shared with CCL4 and CCL5 (RANTES). CCR5 serves as the primary co-receptor for HIV entry, and CCR5 ligands inhibit viral entry through steric hindrance and receptor internalization.
The CR3M clone is specific for CCL3 with no cross-reactivity to CCL4 (MIP-1 beta).
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using stimulated normal human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended protocol: BestProtocols® — Protocol A: Two-step protocol for cytoplasmic intracellular proteins.
Use at 5 µL (0.015 µg) per test (100 µL final volume). Cell number per test: 10⁵–10⁸ cells.

Target Information
MIP-1α (CCL3) and MIP-1β (CCL4) are CC chemokines involved in immune responses to pathogens. They regulate trafficking and activation of macrophages, lymphocytes, and NK cells.
MIP-1α attracts CD8+ lymphocytes, while MIP-1β attracts CD4+ lymphocytes. Both act as chemoattractants for B cells, eosinophils, and dendritic cells, and are active across human and murine hematopoietic systems.
