Product Specific Information

This COX 229 monoclonal antibody recognizes human COX2 (PTGS2) and cross-reacts with mouse and rat COX2.
For optimal performance, use with the Foxp3 / Transcription Factor Staining Buffer Set and protocol.

• Applications Reported: Intracellular staining followed by flow cytometric analysis.
• Applications Tested: Tested by intracellular staining of stimulated normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
• Recommended ≤ 0.125 µg per test (100 µL final volume).
• Suitable for 10⁵–10⁸ cells/test.
• Antibody should be titrated for optimal performance.
• Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.

Target Information

COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis and inflammatory prostaglandin production.
This reaction involves:

1. Cyclooxygenase (COX) activity converting arachidonate to PGG2.
2. Peroxidase activity reducing PGG2 to PGH2.

COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and upregulated under pathological conditions (e.g., cancer, inflammation).
Up-regulation is linked to increased cell adhesion, resistance to apoptosis, and tumor angiogenesis.
COX2 inhibition by NSAIDs (e.g., aspirin) reduces inflammation-related pain and swelling.
Calcitriol (active Vitamin D) naturally inhibits COX2, while glucocorticoids repress PTGS2 gene activity.
