Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control tissue is small intestine.

Target Information

ErbB3 (c-erbB-3/HER-3) is a member of the type I family of growth factor receptors that binds to ligands in the heregulin family.
ErbB3 is over-expressed in various tumors such as those of the prostate, bladder, breast, stomach, pancreas, and colon.
Heregulin and EGF stimulate tyrosine phosphorylation of c-erbB-3 to different extents.
The ErbB3 gene is located on chromosome 12q13 and encodes a transmembrane receptor tyrosine kinase (~160/180 kDa).
ErbB3 forms heterodimers with other EGF receptor family members, leading to activation of pathways involved in cell proliferation and differentiation.
Alternative splicing produces different isoforms, including a secreted form that modulates the activity of the membrane-bound receptor.
