Target Information
This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant).
These belong to the CXC chemokine subfamily, characterized by a single amino acid separating the first two conserved Cys residues. Chemokine binding induces cellular responses related to leukocyte trafficking, including integrin activation, cytoskeletal rearrangement, and chemotactic migration.
Inhibition by Bordetella pertussis toxin indicates coupling with Gi-class heterotrimeric G proteins. Signal transduction may involve enzymes similar to those in other chemokine receptor cascades.
Chemokine-induced desensitization leads to rapid, short-term cellular responses, followed by receptor recycling and restored responsiveness. CXCR3 is highly expressed in effector/memory T cells and T cells in inflamed tissues. Its ligands (IP10, Mig, I-TAC) are produced locally in inflammatory lesions, implicating CXCR3 in immune cell recruitment.
This receptor is a potential target for small molecule antagonists for treating inflammatory diseases. Multiple transcript variants encoding different isoforms exist.
