Product Specific Information

The JES5-16E3 antibody reacts with mouse interleukin-10 (IL-10), an ~18 kDa cytokine also known as Cytokine Synthesis Inhibitory Factor (CSIF). IL-10 is produced by Th2 cells, B1 cells, macrophages, and keratinocytes, and inhibits Th1 cytokine synthesis and T cell proliferation. It acts as a costimulatory signal for mast cells, developing thymocytes, and the Th2 response.

• Capture of mouse IL-10 by ELISA and ELISPOT
• Intracellular staining for flow cytometry
• Immunohistochemistry (IHC)
• Neutralization of IL-10 bioactivity

Applications Tested:
Pre-titrated for intracellular staining and flow cytometric analysis of LPS-stimulated normal human peripheral blood cells. Recommended test size: 5 µL (0.25 µg)/test, suitable for 10⁵–10⁸ cells/test.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production, blocks NF-kappa B activity, and modulates the JAK-STAT signaling pathway. Knockout studies in mice indicate its essential role in intestinal immunoregulation. Mutations in IL10 are associated with increased susceptibility to HIV-1 infection and rheumatoid arthritis. Related diseases include Graft-Versus-Host Disease and HIV-1 infection.
