Product Specific Information

This antibody recognizes the C-terminal region of human ZAP70 protein tyrosine kinase (intracellular antigen).
ZAP70 is susceptible to degradation; therefore, it is recommended to use freshly prepared cell lysates with protease inhibitors to avoid non-specific staining of degradation products.
Western Blot: Reducing conditions are recommended.


Target Information

ZAP-70 is a cytosolic protein tyrosine kinase belonging to the Syk family. It is expressed exclusively in T-cells and natural killer (NK) cells and is essential for T-cell receptor (TCR) activation.
Upon TCR engagement, ZAP70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region between the SH2 and kinase domains.

  • Phosphorylation of tyrosines 315 (Vav binding site) and 319 (Lck binding site) enhances ZAP70-mediated lymphocyte signaling.
  • Phosphorylation of tyrosine 292 attenuates ZAP70 activation and signaling.
