Preparation:
Produced from sera of rabbits immunized with highly pure recombinant murine IL-17A. Anti-murine IL-17A-specific antibody was purified by affinity chromatography employing an immobilized murine IL-17A matrix.

Sandwich ELISA:
To detect murine IL-17A by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Murine IL-17A (500-P265Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of recombinant murine IL-17A.

Western Blot:
To detect murine IL-17A by Western blot analysis, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for recombinant murine IL-17A is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa disulfide-linked homodimer consisting of 136 amino acids, part of the IL-17 family (IL-17A–17F), signaling through IL-17R (CDw217).
Human IL-17A stimulates epithelial, endothelial, and fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2. It enhances ICAM-1 expression and supports proliferation of CD34+ hematopoietic progenitors.
Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating receptor cross-reactivity. IL-17A regulates NF-κB and MAPK activities, stimulates IL-6 and COX-2 expression, and enhances nitric oxide production.
