Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human VEGF-B. Anti-Human VEGF-B-specific antibody was purified by affinity chromatography employing an immobilized Human VEGF-B matrix.

Sandwich ELISA:
To detect Human VEGF-B by sandwich ELISA (using 100 µL/well), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human VEGF-B (500-P267BT) as a detection antibody, allows the detection of at least 2000–4000 pg/mL of Recombinant Human VEGF-B.

Western Blot:
To detect Human VEGF-B by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human VEGF-B is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

Vascular endothelial growth factors (VEGFs) are a family of closely related growth factors with a conserved pattern of eight cysteine residues and shared VEGF receptors. VEGFs stimulate endothelial cells, induce angiogenesis, promote cell migration, increase vascular permeability, and inhibit apoptosis. VEGFB shares structural similarities with VEGF and PIGF and is often co-expressed with VEGF.

There are two alternatively spliced isoforms of VEGFB: VEGFB167 and VEGFB186.

• VEGFB167: highly basic, heparin-binding protein that remains with the cell or extracellular matrix
• VEGFB186: readily secreted

VEGFB stimulates endothelial cell proliferation and binds to VEGFR1 (flt1), but not VEGFR2. It is widely expressed, with high levels in heart, skeletal muscle, and pancreas. VEGFB expression in human primary breast cancers has been associated with lymph node metastasis. The VEGFB gene is located on chromosome 11q13.
